Assembling defined DNA nanostructures with anticancer drugs: a metformin/DNA complex nanoplatform with a synergistic antitumor effect for KRAS-mutated lung cancer therapy

Abstract Herein, a strategy is proposed to simultaneously deliver the small-molecule drug metformin and siRNA with self-assembled DNA nanostructures. The biomedical application of nanostructures highly promising but still in its infancy. as delivery vehicles are conventionally synthesized magnesium-containing buffer. We propose using an anticancer assemble them for synergistic therapy. cargo induces self-assembly into well-defined, uniform nanostructures, producing drug–DNA nanocomplex multiple functionalities cancer Both tile-based origami structures can be assembled metformin. as-prepared metformin/DNA showed high structural thermal stability enzymatic resistance physiological settings. KRAS G12C exerted strong, antitumor effect against KRAS-mutated non-small cell lung (NSCLC) both vitro vivo by suppressing RAS/AKT/mTOR AMPK/AKT/mTOR signaling pathways. current study suggests that assembly complex nanomaterials carefully chosen small molecules key endowing new subsequently expanding their applications multidisciplinary research fields.
